Deadpool introduces Wade Wilson (Ryan Reynolds), a wisecracking mercenary who works in New York City. He falls deeply in love with Vanessa Carlysle (Morena Baccarin), a confident and tough escort. Their idyllic life is cut short when Wade is diagnosed with terminal cancer.
Desperate for a cure, Wade is approached by a shady recruiter offering an experimental procedure that claims to heal his cancer and give him superpowers. He leaves Vanessa, believing she deserves better than to witness his deterioration.
Wade undergoes the procedure at a hidden laboratory run by the sadistic Ajax (Ed Skrein) and his assistant, Angel Dust (Gina Carano). The experiment aims to activate dormant mutant genes in Wade’s body, but it involves excruciating torture.
Though his cancer is cured, the process horribly disfigures Wade, leaving him with burn-like scars all over his body. He develops a powerful regenerative healing factor but escapes the lab, intent on seeking revenge on Ajax for his suffering and disfigurement.
Wade adopts the alter ego of Deadpool, donning a red suit and mask and wielding katanas and pistols. He lives with an elderly blind woman named Al (Leslie Uggams) and enlists the help of his best friend Weasel (T.J. Miller) in tracking down Ajax.
After a series of violent encounters, Deadpool locates Ajax. Their confrontation is interrupted by the X-Men, Colossus (voice by Stefan Kapičić) and Negasonic Teenage Warhead (Brianna Hildebrand), who attempt to convince Deadpool to join them.
Ajax escapes and kidnaps Vanessa. Deadpool convinces Colossus and Negasonic to assist him in rescuing her. They battle Ajax and his forces at a decommissioned Helicarrier in a scrapyard.
